18. Glucagon-like peptide-1 (Active) ELISA KIT

Principles of Assay:

This assay is based on capturing of active GLP-1 from sample by a monoclonal antibody (specific binding to he N-terminal region) followed by tagging this complex with anti GLP-1-alkaline phosphatase detection and finally the quantification of bound will be detected by adding MUP (methyl umbelliferyl phosphate).

Sample Collection and Storage:

Collect blood in ice-cooled Vacutainer EDTA-plasma tubes. Immediately (<30 seconds) add appropriate amount of DPP-IV inhibitor according to manufacturer’s directions. Invert tube to mix and store tubes in ice bath. Centrifuge immediately at 100 xg for 10 minutes in refrigerated centrifuge or place tubes on ice and centrifuge within one hour. Store at 4oC if they will be tested within 3 hours of collection and for longer storage, use a –70oC . Avoid multiple freeze/thaw cycles.

Sensitivity:

The lowest detection range for GLP-1 is 2 pM (in 100ml plasma sample size).

GLP-2 Not detectable
Glucagon Not detectable

Precision:

Within assay precision between 6-9 %CV, and Between assays is from <1 to 13 %CV.
